MGF (Mechano Growth Factor), also known as IGF-1 Ec, is a synthetic 24-amino acid peptide corresponding to the C-terminal E-domain of the insulin-like growth factor 1 Ec splice variant. With the molecular formula C121H200N42O39 and a molecular weight of 2867.26 g/mol, MGF is defined by the amino acid sequence Tyr-Gln-Pro-Pro-Ser-Thr-Asn-Lys-Asn-Thr-Lys-Ser-Gln-Arg-Arg-Lys-Gly-Ser-Thr-Phe-Glu-Glu-Arg-Lys. In native physiology this peptide domain is generated by alternative splicing of the IGF-1 gene and is expressed in a mechano-sensitive manner following tissue strain. Produced via solid-phase peptide synthesis, Mechano Growth Factor is associated with IGF-1 receptor-related signalling pathways in preclinical research.
MGF has been extensively investigated in preclinical growth factor research as the isolated E-domain peptide of the IGF-1 Ec isoform. Unlike mature IGF-1 which consists of the 70-amino acid core protein, Mechano Growth Factor represents the splice-specific C-terminal extension characterised in published studies for its independent signalling properties. In vitro studies in C2C12 myoblast and satellite cell-derived preparations have examined MGF’s interaction with skeletal muscle-derived cell populations, documenting differential roles of the Ec E-peptide relative to mature IGF-1 in myoblast proliferation kinetics and differentiation timing [1]. Rodent model studies have examined the relationship between E-domain peptide presence and downstream cellular responses in skeletal muscle and neural tissue preparations, documenting differential activity across IGF-1 splice variants including the Ec isoform under controlled preclinical conditions [2], making Mechano Growth Factor a key reference compound in IGF-1 splice variant research across skeletal muscle and neural tissue models.
