BPC-157 (CAS 137525-51-0), also known as Body Protection Compound-157, Bepecin, and PL 14736, is a synthetic pentadecapeptide with the molecular formula C₆₂H₉₈N₁₆O₂₂ and a molecular weight of 1419.55 g/mol. Defined by the amino acid sequence Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val, BPC-157 corresponds to a partial sequence of a larger protein (BPC) first isolated from human gastric juice. Notably, the compound is stable across a wide pH range and fully soluble in aqueous media, unusual properties for a peptide of this size that are attributed to its high proportion of proline and charged residues. Produced via solid-phase peptide synthesis, BPC-157 is associated with nitric oxide (NO) signalling, VEGF expression, and FAK-paxillin pathway.
BPC-157 is one of the most extensively referenced cytoprotective peptides, with studies spanning gastrointestinal, musculoskeletal, cardiovascular, and central nervous system research contexts. In vitro studies have examined its interaction with endothelial cell migration, fibroblast proliferation, and nitric oxide synthase (NOS) system modulation, documenting activation of the VEGFR2-Akt-eNOS and Src-Caveolin-1-eNOS signalling axes in vascular endothelial cells [1]. Tendon fibroblast studies have additionally documented BPC-157 upregulation of growth hormone receptor (GHR) expression, a receptor that signals canonically through the JAK2-STAT pathway [2]. In vivo rodent studies have investigated BPC-157 across gastrointestinal ulcer models, tendon and ligament transection models, and peripheral nerve injury paradigms, with research examining angiogenesis and new-vessel and granulation-tissue formation in both gastrointestinal mucosa and musculoskeletal tissue [3][4], alongside collagen deposition kinetics and, in a separate body of central nervous system work, dopaminergic and serotonergic system dynamics, following both systemic and local administration.
