Sermorelin (CAS 86168-78-7), also known as GRF(1-29), is a synthetic 29-amino acid peptide corresponding to the first 29 residues of endogenous human growth hormone-releasing hormone (GHRH). With the molecular formula C149H246N44O42S and a molecular weight of 3357.93 g/mol, Sermorelin acts at the GHRH receptor (GHRH-R) and is structurally and mechanistically distinct from the ghrelin receptor (GHS-R1a) agonists in the secretagogue class. Sermorelin is one of the most extensively studied GHRH analogues in preclinical neuroendocrinology. As a GHRH-R agonist, it engages a fundamentally different receptor system to the ghrelin-mimetic secretagogues such as GHRP-6 and Ipamorelin, making it an established comparator compound in studies examining the dual regulatory control of somatotroph function. Published in vitro research in rat anterior pituitary cell preparations has investigated Sermorelin/GRF(1-29)-mediated cyclic AMP accumulation and downstream somatotroph signalling, documenting inhibitory modulation of this response by somatostatin at the level of GHRH-R activation [1]. In vivo rodent studies have examined GH secretion patterns following Sermorelin/GRF(1-29) administration, with particular focus on ultradian pulse amplitude modulation and the interplay between GHRH and somatostatin tone within the hypothalamic-pituitary axis [2].
