What is FOXO4-DRI (Proxofim)?
FOXO4-DRI is a synthetic peptide built to clear senescent cells. Those are worn-out cells that stop dividing but refuse to clear away, and they build up in tissue over time. The peptide is copied from a stretch of a human protein called FOXO4, then rebuilt in two ways. The residue order is reversed, and every residue is the mirror-image D form rather than the L form found in the body. That is what retro-inverso means. The point of the rebuild is that the enzymes which chop up ordinary peptides do not recognize the mirror image. A second, positively charged sequence is attached to help it cross cell membranes.

Yes, and it is on the Published Research tab. A 2023 study in Circulation cleared senescent cells by four different routes, one of which was this peptide, and reported that doing so worsened a lung blood-vessel disorder in mice and rats. Its authors conclude that eliminating senescent pulmonary endothelial cells may worsen pulmonary hemodynamics, and the journal ran an editorial on the finding in the same issue. Separately, a study in human cartilage cells removed the senescent cells successfully but did not improve the cartilage those cells went on to produce.
