What is GHRP-2?

GHRP-2 (Growth Hormone Releasing Peptide-2), also referred to as Pralmorelin and KP-102, is an synthetic Growth Hormone Secretagogue with the molecular formula C₄₅H₅₅N₉O₆ and molecular weight 817.99 g/mol. GHRP-2 features a D-2-naphthylalanine side-chain at the position 2 of the amino acid chain, a characteristic feature that is considered critical for ligand recognition by the ghrelin receptor (GHS-R1a). Produced via solid-phase peptide synthesis, Pralmorelin (CAS 158861-67-7) is among the most widely referenced second-generation secretagogues in preclinical growth hormone research.
GHRP-2 has been extensively investigated in preclinical endocrinology and neuroendocrine pharmacology. In vitro studies conducted on primary cultured ovine and rat pituitary somatotroph cell lines have examined the signalling process of GHRP-2/Pralmorelin in the activation of its GHS-R1a receptor through intracellular production of cAMP, voltage-dependent activation of Ca+ channels, and involvement of distinct PKC and PKA pathways in the stimulation of GH secretion [1]. In vivo studies in rodents have examined the role of GHRP-2/KP-102 within the somatotropic pathway, with evidence suggesting that GHRP-2 acts synergistically with GHRH mechanisms during pulsatile GH secretion [2]. The replacement of D-Tryptophan residue present in previous generation peptide GHRP-6 with D-2-Naphthylalanine marks the difference in structure between GHRP-2 [3], and GHRP-6 and has made GHRP-2 a key reference compound in comparative studies within the growth hormone secretagogues class.
