What is PAL-GHK?

PAL-GHK (Palmitoyl Tripeptide-1) is a synthetic lipopeptide formed by conjugating palmitic acid to the copper-binding tripeptide GHK (Gly-His-Lys), typically supplied without the coordinated copper ion present in the parent GHK-Cu complex. GHK-Cu, first isolated from human plasma, has been shown in vitro to stimulate collagen synthesis in fibroblast cultures at low, physiologically relevant concentrations, consistent with GHK's proposed role as a wound-healing signal released during collagen breakdown [1]. The palmitoyl modification is intended to increase the hydrophobicity of the GHK sequence, and a 2024 review of GHK-based anti-wrinkle peptides reports that this raises PAL-GHK's calculated partition coefficient and its measured skin permeation relative to both unmodified GHK and GHK-Cu [2]. The same review is explicit that this permeability rationale, rather than dedicated bioactivity data, is the current basis for PAL-GHK's cosmetic use, noting that cellular or clinical studies evaluating its own anti-wrinkle activity are still lacking in the published literature [2].
