Colivelin is a synthetic hybrid neuropeptide with an approximate molecular weight of 3000 g/mol, constructed as a fusion of two independently characterised neuroprotective sequences. Its N-terminal domain consists of the nine-amino acid core peptide of activity-dependent neurotrophic factor (ADNF9: Ser-Ala-Leu-Leu-Arg-Ser-Ile-Pro-Ala), C-terminally fused to AGA-(C8R)HNG17, a potent 17-amino acid derivative of Humanin incorporating stabilising amino acid substitutions at multiple positions. This chimeric design combines two structurally and mechanistically independent neuroprotective signalling domains within a single peptide chain. Produced via solid-phase peptide synthesis, Colivelin activates two distinct intracellular signalling pathways.
Colivelin is designed hybrid construct combining two independent neuroprotective signalling mechanisms. Published research has characterised the ADNF domain as triggering a Ca2+/calmodulin-dependent protein kinase IV (CaMKIV) cascade and the Humanin-derived domain as signalling through a STAT3-dependent pathway, with both mechanisms contributing to the compound’s neuroprotective activity in neuronal cell preparations. In vitro studies have documented that Colivelin suppresses neuronal cell death induced by overexpressed familial Alzheimer’s disease genes and amyloid-beta at femtomolar concentrations and have characterised this neuroprotection as dependent on both an ADNF-triggered CaMKIV pathway and a Humanin-domain-triggered STAT3 pathway [1][2]. In vivo rodent studies documented that central administration of Colivelin suppressed spatial working memory impairment induced by repetitive amyloid-beta injection and antagonised neuronal loss in the hippocampal CA1 region, while peripheral administration was active in a cholinergic amnesia model, indicating neuroprotective and pro-cognitive activity following both central and peripheral administration [2], making Colivelin a reference compound in preclinical neuroscience research examining rationally designed hybrid neuroprotective peptides and dual-pathway (CaMKIV/STAT3) cytoprotective signalling in preclinical models.
