DSIP (CAS 62568-57-4), also known as Delta Sleep-Inducing Peptide and by the INN Emideltide, is a synthetic nonapeptide with the molecular formula C35H48N10O15 and a molecular weight of 848.82 g/mol. Defined by the amino acid sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, DSIP is classified as an amphiphilic neuropeptide first isolated from rabbit cerebral venous blood in 1977. The N-terminal tryptophan residue provides a characteristic UV absorbance signature used for laboratory identification. Produced via solid-phase peptide synthesis, Delta Sleep-Inducing Peptide is associated with neuroendocrine and circadian signalling pathways.

DSIP has been extensively investigated across a broad range of preclinical neuroendocrine research contexts. The compound has been identified in both free and bound forms within the hypothalamus, limbic system, pituitary, and peripheral tissues, and has been documented as co-localising with multiple peptide and non-peptide mediators including ACTH, melanocyte-stimulating hormone (MSH), and thyroid-stimulating hormone (TSH). Studies examining DSIP immunoreactivity in human pituitary tissue preparations have characterised DSIP’s interaction with neuroendocrine cell populations, documenting its co-localisation with corticotropin-like intermediate lobe peptide (CLIP) in anterior pituitary corticotroph cells and residual intermediate lobe cell populations, establishing a neuroendocrine co-expression pattern within hypophyseal tissue [1]. In vivo rodent studies have examined the relationship between DSIP administration and sleep architecture, with research documenting increases in electroencephalographic slow-wave activity and characterising the interplay between DSIP and endogenous hormonal rhythms, with evidence that endogenous DSIP plays a physiological role in slow-wave sleep and circadian neuroendocrine regulation established through antiserum microinjection studies in rats [2], making DSIP a reference compound in neuropeptide research examining the neuroendocrine and circadian signalling correlates of sleep architecture.
