Crystagen is a synthetic tripeptide bioregulator with the sequence Glu-Asp-Pro (EDP), corresponding to the molecular formula C₁₄H₂₁N₃O₈ and a free-acid molecular weight of approximately 359.33 g/mol. It belongs to the Khavinson family of ultrashort peptide bioregulators developed at the Saint Petersburg Institute of Bioregulation and Gerontology. EDP is one of the active short-peptide constituents of the thymus-derived polypeptide complex Thymalin. Produced via solid-phase peptide synthesis, Crystagen is classified as an immune-system (thymic) bioregulator.
In preclinical research models, Crystagen is studied for its capacity to modulate immune-cell activity. The immunomodulatory profile of the thymic complex to which EDP belongs has been directly characterised in vitro: in the human THP-1 monocyte/macrophage line, Thymalin and related Khavinson thymic peptides modulated proliferative activity and inflammatory cytokine pathways, consistent with a regulatory (rather than purely stimulatory) action on innate immune cells [1]. At the mechanistic level, and consistent with the proposed direct-to-DNA mode of action, fluorescently labelled peptides of this ultrashort class penetrate the cell nucleus and bind genomic DNA in a sequence-selective manner, providing a structural basis for peptide-directed modulation of tissue-specific gene expression [2]. Within the immune compartment, EDP-type thymic peptides are studied for effects on thymocyte differentiation and proliferation and on macrophage cytokine output, including in aged models where thymic involution is examined, making Crystagen a reference compound in preclinical immunology research examining thymic function, macrophage activation, and age-related immune decline.
