PEG-MGF (PEGylated Mechano Growth Factor) is a synthetically modified 24-amino acid peptide derived from the IGF-1 Ec splice variant, covalently conjugated to a polyethylene glycol (PEG) moiety. The core peptide corresponds to the C-terminal E-domain of Mechano Growth Factor, with an approximate molecular weight of 4500 g/mol inclusive of the PEG chain. PEGylation is a well-established chemical modification strategy employed to alter the physicochemical characteristics of the native peptide, including its susceptibility to enzymatic degradation. Produced via solid-phase peptide synthesis with subsequent PEG conjugation, PEG-MGF is associated with IGF-1 receptor-mediated signalling pathways in preclinical research.
PEGylated Mechano Growth Factor has been extensively investigated in preclinical cell biology and tissue physiology research, with particular focus on satellite cell populations and their role in skeletal muscle adaptation. In vitro studies in C2C12 myoblast cell lines have examined the compound’s interaction with the IGF-1R, characterising differential contributions of the Ec E-peptide to myoblast proliferation and differentiation relative to mature IGF-1, with downstream signalling activity documented across MAPK/ERK-mediated mitogenic pathways [1]. In vivo studies have explored the pharmacokinetic implications of PEGylation on MGF, comparing circulating half-life and tissue distribution profiles between PEG-conjugated and non-conjugated forms. Rodent cardiovascular models have also examined the E-domain peptide’s interaction with cardiac tissue, with published work documenting MGF E-peptide activity in rat cardiomyocyte preparations and in infarcted myocardium in vivo [2], an area that has made PEG-MGF a key subject of preclinical investigation beyond the skeletal muscle axis.
