TB-500 (CAS 885340-08-9), the synthetic form of Thymosin Beta-4 (Tβ4), is a 43-amino acid peptide with the molecular formula C₂₁₂H₃₅₀N₅₆O₇₈S and a molecular weight of 4963.44 g/mol. Its full sequence (Ac-Ser-Asp-Lys-Pro-Asp-Met-Ala-Glu-Ile-Glu-Lys-Phe-Asp-Lys-Ser-Lys-Leu-Lys-Lys-Thr-Glu-Thr-Gln-Glu-Lys-Asn-Pro-Leu-Pro-Ser-Lys-Glu-Thr-Ile-Glu-Gln-Glu-Lys-Gln-Ala-Gly-Glu-Ser) features an N-terminal acetylation and contains the critical LKKTETQ actin-binding motif at positions 17-23. TB-500 belongs to a highly conserved family of 16 beta-thymosin isoforms and is one of the most abundant intracellular peptides in mammalian cells. Produced via solid-phase peptide synthesis, TB-500 functions as a G-actin sequestering protein.

TB-500 is central to preclinical cytoskeletal biology and tissue remodelling research. Its primary mechanism centres on the sequestration of monomeric globular actin (G-actin), regulating the equilibrium between G-actin monomers and polymerised filamentous actin (F-actin), a balance governing cell migration, morphogenesis, and cytoskeletal reorganisation. The conserved central LKKTET motif (positions 17-22) is essential for actin binding, but mutational mapping has shown that the isolated motif alone binds G-actin only weakly; high-affinity sequestration depends on additional contacts contributed by regions flanking the motif across the full-length peptide [1]. In vitro studies have examined TB-500 in endothelial cell, fibroblast, and keratinocyte preparations, documenting enhanced endothelial cell differentiation and capillary-like tube formation on reconstituted basement membrane [2], alongside directional cell migration and extracellular matrix (ECM) remodelling through matrix metalloproteinase (MMP) expression. In vivo rodent studies have investigated TB-500 in dermal wound, corneal injury, and cardiac ischaemia models, reporting accelerated full-thickness dermal wound closure accompanied by increased angiogenesis and collagen deposition [3], findings that span the neovascularisation, collagen-deposition, and inflammatory-to-proliferative phases of the tissue remodelling cascade.
