What is Orexin B?
Orexin B is a chain of 28 amino acids made by nerve cells in the hypothalamus. It is also called hypocretin-2, and most published work uses that name. It comes from the same starting protein as Orexin A, which is sold separately here, but the two are cut into different pieces and behave differently. Orexin B is a straight chain with no internal links, where Orexin A is folded and held by two bridges. There is also a laboratory-made version of Orexin B with two links swapped, and much of the work described as "OX2-selective" uses that version rather than this one.

Is Orexin B selective for the OX2 receptor?+−
Not in the way the phrase is usually meant. The 400-fold OX2R selectivity widely quoted in this area was measured on [Ala11, D-Leu15]-orexin B, an engineered analogue carrying two residue substitutions, and the paper reporting it does not give a selectivity ratio for the native peptide. Published work on native Orexin B in rat brain slices reports one effect mediated by OX2 receptors and a second that runs through neither orexin receptor. Is Orexin B the same as Orexin A?+−
No. They are two peptides cut from the same precursor protein. Orexin A is 33 residues, folded, with two internal disulfide bridges and a modified N-terminus; Orexin B is 28 residues and linear. Published work reports different effects for the two in the same brain region, and in a wake-promotion comparison Orexin A was the more potent. Findings for one are not findings for the other.
